Dr. Weinstein’s work provides insights into the roles of genetic factors, specific environmental factors, and dietary factors in human cancer causation, with the ultimate goal of cancer prevention. The methods employed include cell culture systems, DNA transfection, retrovirus-derived vectors, a variety of recombinant DNA methods, RNAi, microarray analysis, rodent models of chemical carcinogenesis, genetically engineered mice, and immunohistochemistry studies of human tumor tissues. His studies have revealed abnormalities in the internal circuitry of cancer cells that play a role in their uncontrolled proliferation. In related studies he elucidated mechanisms by which EGCG, a major constituent of green tea, and other naturally occurring or synthetic compounds, inhibit growth and induce apoptosis in human cancer cells. In recent studies he used mouse genetics to demonstrate that a gene designated PKCI/HINT1, originally cloned in his laboratory, is a tumor suppressor gene. These findings suggest novel strategies and agents for cancer prevention and treatment.
